Comparative test of methods applied for the measurement of cell concentration in boar semen

Abstract:

In swine breeding artificial insemination is a widely used technology. For efficiency the semen taken from one boar is divided to several insemination doses, so that the number of cells provided in each insemination dose shall result in successful fertilization. For this purpose a method determining cell concentration is necessary, that practically shall be easy to use, simple and exact.

The aim of our study was to compare the most frequently used method for the measurement of cell concentration by spectrophotometer with Makler chamber and CASA (Computer Assisted Semen Analysis). For the study the semen sample of 84 boars (n=349) were used. The concentration of 179 semen samples were examined by spectrophotometer and CASA, the other 170 semen sample by spectrophotometer and Makler chamber.

From the absorbance value (ABSZ) measured by spectrophotometer the concentration could be calculated by a linear equation created previously. This equation for calculating concentration was not applicable at 2,58% of the semen samples (n=9), where the ABSZ value was lower than 0,40, as it resulted a concentration with negative sign concentration and dilution.

In the ABSZ range between 0.51-0.69 the difference in concentrations (n=35) was lower than 10% by spectrophotometer/CASA and by spectrophotometer/Makler chamber. In ABSZ ranges lower (<0,51) and higher (>0,69) the difference increased, that resulted in multiple differences in the concentration at lower and higher ABSZ values. Although CASA measurement consequently provided higher concentrations (by 28,0%) compared by Makler chamber that is considered to be the “gold standard”, but in both measurement methods not linear but exponential connection was observed between ABSZ values and the concentration values measured by the two measurement methods.

Based on these measurements a new equation applying exponential correlation on logarithmic base was created that provides a more exact concentration calculation applicable in all ABSZ ranges.
